Biography
Kathleen Doane, PhD, received her PhD in cell and developmental biology from Rutgers, The State University of New Jersey and the University of Medicine and Dentistry of New Jersey. She did her postdoctoral work at Tufts Medical School. Her research interests concentrate on the interaction of cells with the extracellular matrix using matrix receptors. Dr. Doane came to Geisinger Commonwealth from Northeastern Ohio Universities College of Medicine.
Research interests
Dr. Doane’s major research interests concentrate on the interaction of cells with the extracellular matrix using matrix receptors. Her major project elucidates the cell-matrix interactions important in heart failure fibroblasts. This NIH funded project (1R15HL079969) is examining the role of collagen VI and matrix receptor signaling in transgenic type VI collagen knockout mice. Interaction of fibroblasts with type VI collagen has been shown to cause these cells to differentiate into myofibroblasts, a wound healing phenotype that is hyperproliferative and hypersecretory. We have demonstrated an increase in type VI collagen following myocardial infarction and subsequent to administration of isoproterenol, a synthetic catecholamine.
An increase in markers for myofibroblasts indicates a possible correlation between the presence of these cells and type VI collagen during a disease/wound healing state. Our hypothesis is that cells subjected to abnormal conditions begin to secrete type VI collagen, which causes excess myofibroblast differentiation. This may lead to excessive fibrosis during wound healing due to the hypersecretion by these cells, and development of a fibrotic scar due to excess proliferation. Control of the deposition of the collagen or blockade of the signaling pathway leading to myofibroblast differentiation could result in new treatment paradigms for heart disease.
Other projects:
Using breast cancer cell lines to study the interaction of integrins and urokinase plasminogen activator receptor at different levels of invasiveness (collaboration with Akron General Medical Center).
Development of a liquid-crystal-based pathogen detection system. There are two licensees and several patents covering this technology (collaboration with Northeastern Ohio Universities College of Medicine, Kent State University and Pathogen Systems Inc.).
Analysis of phenotypic alterations in synovial fibroblasts following metal challenge as a model for development of joint failure following arthroplasty (collaboration with Summa Health Care Systems).
Gene therapy by nanosphere delivery using a kidney hypertension model (collaboration with University of Akron and Walsh University).
